Objective: The study aimed to investigate the antimicrobial profile for the distribution and the possible relationship outcome of integrons related to meropenem and imipenem resistance in P. aeruginosa isolates.
Methods: A total of 106 P. aeruginosa isolates were identified by randomly collected samples from clinical isolates in this cross-sectional study was conducted over 12-month study duration from October 2023 to September 2024. Identification was done by standard conventional microbiological and biochemical screening using Pseudomonas isolation agar and MacConkey’s agar medium. Molecular confirmation was done by species-specific 16S rRNA gene profiling. Antibiotic susceptibility testing was performed using the minimum inhibitory concentration method following CLSI guidelines. Statistical analysis of carbapenem resistance patterns was assessed by Kandall’s tau-b test using SPSS 20 software. For molecular analysis, DNA was extracted via the heat-lysis method, and to identify the strains carrying integrons, the conserved regions of the integron-encoded integrase gene Intl1 were amplified by using polymerase chain reaction (PCR) with specific primers, and the bands were confirmed by gel electrophoresis, which was then visualized under a UV - transilluminator.
Results:The drug susceptibility of MIC findings of meropenem and imipenem showed 92% susceptibility in 1?g/ml concentration; however, only 13 % strains showed resistance in 4 ?g/ml of concentration. The p- value of susceptibility showed a significant Kandall’s tau-b test value of 1. However, Class 1 integron genes were detected in 10% of isolates. Integron presence was strongly associated with multidrug resistance.
Conclusion:The prevalence of integron genes among clinical isolates of P. aeruginosa highlights their central role in spreading antibiotic resistance and their close relation with carbapenem drug resistance. Responsible antibiotic use and regular surveillance in the community are crucial to curb the spread of multidrug-resistant bacteria for a successful treatment outcome.
